Abstract:ObjectiveTo determine the amino acids in root of Rumex gmelini with precolumn derivatization. MethodsSixteen kinds of amino acids were determined in root of Rumex gmelini by pre-column of RPHPLC. HPLC analysis was performed on Dikma C18 column with mixture of 0.05mol/L sodium acetate to contain 1% N,N-dimethylformamide (A) and acetonitrile-water (1∶1) (B) as mobile phase in gradient mode. The flow rate was 1.0ml/min. The detection wavelength was 360nm and the column temperature was 40℃. ResultsThe molar concentrations of amino acids and peak areas achieved a nice linear relationship (r was higher than 0.999). The recoveries of sixteen kinds of amino acids were 94.7%~112.9%. ConclusionThe method is accurate and repeatable for the determination of amino acids in Rumex gmelini.
